In this study, 874 unique lentiviral vector integration sites in human HaCaT keratinocytes after long-term culture were identified and analyzed with the online tool GTSG-QuickMap and SPSS software.
The data indicated that lentiviral vectors showed integration site preferences for genes and gene-rich regions.
This study will likely assist in determining the relative risks of the lentiviral vector system and in the design of a safe lentiviral vector system in the gene therapy of skin diseases.

In this study, the barrier activity of an insulator element from the human ankyrin-1 gene was analyzed in a lentiviral vector carrying an antisickling human β-globin gene. The presence of the Ankyrin insulator element did not change transgene expression in human hematopoietic cells in short-term erythroid culture or in vivo in primary murine transplants. However, analysis in secondary recipients showed that the lentiviral vector with the Ankyrin element preserved transgene expression, whereas expression from the vector lacking the Ankyrin insulator decreased in secondary recipients.

The authors have constructed several lentiviral vector to express the factor VIII under the control of different promoters : MND, EF1 and glycoprotein-1bα (G1B). Their results show that only the G1B-FVIII vector achieves long-term and efficient expression, resulting in partial correction of hemophilia A.
